Viking's VK2735 Holds Up to 97% of Weight Loss on Monthly and Biweekly Maintenance Dosing

Viking Therapeutics has reported positive topline results from a maintenance study of VK2735, its dual GLP-1/GIP receptor agonist, showing that patients who lose weight on weekly injections can keep nearly all of it after stepping down to monthly or every-other-week dosing. What Happened

The randomized, double-blind, placebo-controlled VK2735-102 study enrolled approximately 180 adults with obesity. It ran in two phases:

• Induction (21 weeks): Participants received once-weekly subcutaneous VK2735 or placebo. Mean weight loss ranged from approximately 16% to 19% versus approximately 0% for placebo, with no plateau through week 21.
• Maintenance (12 weeks): VK2735 participants were re-randomized to every-other-week or monthly regimens, or placebo.

The headline maintenance numbers:

• Every-other-week dosing preserved up to 97% of induction weight loss (p<0.0001), versus 61% for placebo.
• Monthly dosing preserved up to 90% (p=0.0002).
• An exploratory cohort that stayed on weekly dosing reached 21.3% mean weight loss at week 33, still progressive.
• In combined treatment groups, 98% of participants lost at least 5% of body weight and 65% lost at least 15%, versus 13% and 0% for placebo.

Tolerability during maintenance was excellent: gastrointestinal adverse event rates were not meaningfully different from placebo, and discontinuations due to adverse events were low.
